The MassPREP online desalting column can efficiently desalt proteins prior to LC-MS analysis. Non-volatile salts (e.g. NaCl) can suppress the ionization of intact proteins, resulting in poor detection sensitivity. It is therefore important to eliminate or significantly minimize the entry of these compounds into the mass analyzer. The reversed-phase phenyl material contained in the MassPREP online column successfully 'traps' the proteins, allowing the salts to be washed into the waste prior to elution of the proteins into the mass spectrometer. With an optimized LC-MS method, cycle times as short as 4 minutes (for intact antibodies) and 10 minutes (for reduced antibodies) can be achieved.
